CD56dim and CD56bright cells were distinguished by appropriate gating in the CD56+ region. Whole-blood aliquots with appropriate MAbs were incubated in the dark at room temperature for 20 min. mononuclear cells (PBMC) were isolated from heparinized whole blood by Ficoll-Hypaque (Pharmacia, Uppsala, Sweden) density gradient centrifugation. Freshly isolated Ficoll-purified PBMC were adjusted to 1 × 107 cells per milliliter in complete medium and were then diluted serially at 40:1, 20:1, 10:1, and 5:1 effector-to-target (E:T) ratios. The PBMC were placed into 96-well round-bottom microtiter plates and incubated with radiolabeled K562 cells. K562 cells were labeled with 100 μCi·10−6 cells of sodium 51chromate (51Cr; ICN Biomedicals, Irvine, CA) over a 1-h period in a shaking waterbath at 37 °C.
